Zonulin (+ Zonulin Family Peptides)

Zonulin is the protein that controls the tight junctions of your gut lining. Those junctions are what keep the good stuff in your bloodstream and the bad stuff out. When Zonulin is elevated, those junctions open when they shouldn't. Bacteria, food particles, and other triggers can cross into circulation, setting off inflammation, food sensitivities, and immune reactions. This is what leaky gut actually means, and this is the first test to measure it with a real biomarker.

Histamine

Histamine is a chemical messenger your immune system produces. In normal amounts it does important jobs. When it accumulates, it drives a wide range of symptoms that look random but aren't. A compromised gut lining increases histamine production while simultaneously reducing your body's ability to clear it. That combination is why so many people with gut issues end up reactive to foods, environments, and stress in ways that feel completely unpredictable.

DAO (Diamine Oxidase)

DAO is the enzyme responsible for breaking down histamine in your gut. When your gut is inflamed, DAO production often drops, which means histamine builds up regardless of how careful you are with your diet. This is the piece most people are missing. It's not always that you're eating too much histamine. It's often that your body can't clear what's already there. And here's where the thyroid connection comes in: hypothyroidism suppresses DAO production, which means thyroid dysfunction can make histamine intolerance significantly worse even when diet is clean.

Low DAO is often why a low-histamine diet helps some but never fully resolves symptoms on its own.

Histamine : DAO Ratio

This is the most useful number on the panel. A high ratio means there's more histamine circulating than your DAO can manage. It tells us whether the problem is high histamine, low DAO, or both. That distinction changes what comes next.

The ratio is what turns a data point into a direction.

Thyroid function and histamine clearance are directly linked. Hypothyroidism, including subclinical cases and Hashimoto's, suppresses DAO production, which means your body clears histamine less efficiently even before diet enters the picture. Is this just one test? Why not a bigger panel?

More markers isn't always more clarity. Zonulin, DAO, and Histamine are the three markers that speak most directly to leaky gut and your body's ability to clear histamine. The test also calculates the Histamine:DAO ratio, which is often the most useful number of all. Paired with the educational series that walks you through how to read each one, that picture is specific enough to tell you where to focus.

What if my results come back normal?

That's still useful information. Normal results in the presence of significant symptoms usually point somewhere worth investigating. The educational series will help you understand what normal actually means for each marker and what to consider next.

Does insurance cover this?

Do I need a doctor's order?
